Structure-activity work shows that fatty acid length, linker chemistry and the position of acylation all influence albumin affinity and receptor potency. Plasma protein binding exceeds 99 percent, which restricts distribution and slows renal clearance. Degradation proceeds largely through general proteolysis and fatty acid oxidation rather than cytochrome P450 metabolism, so exposure to common oxidative drug interactions is limited. Whether these clearance routes vary meaningfully between individuals is not fully established.
The molecule is a synthetic 39-amino-acid peptide whose backbone derives from the sequence of human glucose-dependent insulinotropic polypeptide, with several substitutions that raise metabolic stability and shift receptor preference. A C20 fatty diacid is attached through a short linker to a lysine side chain, a modification that increases binding to serum albumin. The reported monoisotopic mass is approximately 4813 Da. Near neutral pH the peptide carries a net negative charge, and the lipid tail makes the molecule markedly more hydrophobic than the unmodified parent sequence.
Dual agonism at the GIP and GLP-1 receptors underlies the observed pharmacology. Activation of GLP-1 receptors raises glucose-dependent insulin release, lowers glucagon secretion, slows gastric emptying and reduces appetite. GIP receptor activation contributes additional effects on adipose tissue and on energy balance, and the combined action on appetite appears larger than either pathway alone in animal models. Signalling bias and the relative contribution of each receptor arm to weight-related effects remain areas of active investigation.
Identity and purity are established with reversed-phase high-performance liquid chromatography, often paired with mass spectrometry for confirmation of the expected mass. Peptide mapping after enzymatic digestion verifies the primary sequence and detects substitutions. Size-exclusion chromatography quantifies aggregates and fragments, which are the impurities most often tracked for peptides of this size. Residual solvents, counterions, and water content fall under separate tests described in pharmacopeial chapters. Circular dichroism or nuclear magnetic resonance may be used in research settings to probe secondary structure, though such methods are less common in routine release testing.
Peptide active ingredients of this type are typically supplied as lyophilized powder because the dry form resists hydrolysis during transport. The material is hygroscopic, so vials are usually equilibrated to room temperature before opening to avoid condensation on the solid. Repeated freeze-thaw cycles can promote aggregation and are generally avoided by aliquoting stock into single-use portions. Personnel handling the powder work in controlled environments to limit inhalation of fine particles. Written procedures usually specify these steps rather than leaving them to individual judgment.

After subcutaneous injection, absorption is gradual, and peak plasma levels are generally reached within one to three days. Albumin binding extends the apparent half-life to roughly five days, which supports a weekly administration schedule. Metabolism proceeds mainly through proteolytic cleavage of the peptide backbone and beta-oxidation of the fatty acid chain, rather than through cytochrome P450 pathways. Eliminated fragments are largely recycled through general protein turnover, and excretion of intact drug in urine is minimal. These properties distinguish the molecule from short-acting incretin mimetics.
Tirzepatide is a synthetic peptide of 39 amino acids engineered from the native glucose-dependent insulinotropic polypeptide sequence. Its structure incorporates several non-natural residues and a C-terminal segment derived from glucagon-like peptide-1, together with a C20 fatty diacid moiety attached through a linker. The lipophilic side chain promotes binding to serum albumin, which slows renal clearance after administration. The compound is classified as a dual incretin receptor agonist and is supplied as a lyophilized powder for reconstitution or as a preformulated solution, depending on the presentation.
The peptide activates two G protein-coupled receptors, GIPR and GLP-1R. Binding triggers adenylyl cyclase activity and raises intracellular cyclic AMP in pancreatic beta cells, which potentiates insulin release when glucose is elevated. Signaling in the central nervous system is associated with reduced appetite and lower energy intake, while effects on gastric emptying and glucagon secretion are also reported. Because activity at both receptors is retained, the pharmacological profile is often described as incretin-based rather than selective for a single receptor.
Receptor activation by tirzepatide raises intracellular cyclic AMP through Gs-coupled signalling at both targets. At the GLP-1 receptor the downstream effect includes glucose-dependent insulin release, suppressed glucagon secretion, delayed gastric emptying, and reduced appetite signalling in the hypothalamus. GIP receptor engagement adds insulinotropic activity and appears to influence lipid handling in adipose tissue. Because both receptors are stimulated at the same time, the pharmacological profile differs from that of selective GLP-1 receptor agonists, and the relative contribution of each arm remains an area of active investigation.
Clinical development proceeded through large phase 3 programmes in type 2 diabetes and in obesity or overweight with at least one weight-related comorbidity. Regulatory approvals followed in several jurisdictions for both indications. Weekly subcutaneous dosing reflects an elimination half-life of roughly five days. Open questions include the durability of metabolic effects after treatment stops, long-term cardiovascular and hepatic outcomes beyond completed trials, and whether the dual mechanism confers benefits independent of total receptor occupancy. Published literature continues to expand on these points. Substantial uncertainty remains about interindividual variability in response.
Tirzepatide is a synthetic linear peptide of 39 amino acids that acts as a dual agonist at the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. Its sequence derives from native GIP but incorporates non-natural residues and a C20 fatty diacid moiety linked to a lysine side chain. The lipophilic chain promotes albumin binding, which slows renal clearance and extends circulation time. The unmodified peptide has a molecular formula of C225H348N48O68 and a molecular mass near 4,813 daltons.
== Taxonomy == This species was first formally described in 1753 by Carl Linnaeus who gave it the name Cassia occidentalis in Species Plantarum from specimens collected in Jamaica. In 1829, Link transferred the species to the genus Senna as S. occidentalis in his Handbuch zur Erkennung der nutzbarsten und am häufigsten vorkommenden Gewachse. The specific epithet (occidentalis) means "western".
=== Single-molecule force spectroscopy === Single molecule techniques such as optical tweezers and AFM have been used to understand protein folding mechanisms of isolated proteins as well as proteins with chaperones. Optical tweezers have been used to stretch single protein molecules from their C- and N-termini and unfold them to allow study of the subsequent refolding. The technique allows the measurement of folding rates at single-molecule level; for example, optical tweezers have been recently applied to study folding and unfolding of proteins involved in blood coagulation. von Willebrand factor (vWF) is a protein with an essential role in blood clot formation process. It was discovered – using single molecule optical tweezers measurement – that calcium-bound vWF acts as a shear force sensor in the blood. Shear force leads to unfolding of the A2 domain of vWF, whose refolding rate is dramatically enhanced in the presence of calcium. It has also been shown that the simple src SH3 domain accesses multiple unfolding pathways under force.
In the Philippines, 7-Eleven was run by the Philippine Seven Corporation (PSC). Its first store, located at the corner of EDSA and Kamias Road in Quezon City, opened on February 29, 1984. On July 28, 1988, PSC transferred the Philippine area license to operate 7-Eleven stores to its affiliate, Phil-Seven Properties Corporation ("PSPC"), together with some of its store properties. In exchange thereof, PSC received 47% of PSPC stock as payment. On May 2, 1996, the stockholders of both PSC and PSPC approved the merger of the two companies to advance PSC group's expansion. On October 30, 1996, Securities and Exchange Commission approved the merger and PSPC was then absorbed by PSC as the surviving entity. In 2000, President Chain Store Corporation (PCSC) of Taiwan, also a licensee of 7-Eleven, purchased the majority shares of PSC and thus formed a strategic alliance for the convenience store industry within the area. In February 2009, 7-Eleven signed a non-exclusive contract with Chevron Philippines to open its stores in selected Caltex gas stations nationwide. In 2012, they opened their first store outside of Luzon in Cebu City, which soon expanded to the other parts of Cebu as well as its neighboring provinces. It was followed with the branch openings in Bacolod in 2013, Iloilo City in 2014, Davao City and Cagayan de Oro in 2015. The number of stores eventually spread from these major cities to smaller towns and provinces near them.
=== China === China is the main supplier of wolfberry products globally, with 2023 production derived from a cultivation area of 325,000 mu – about 21,667 ha (53,540 acres) – creating a national wolfberry economy of 29 billion yuan (approximately US $4.2 billion). Some 61% of commercially produced wolfberries in China comes from L. barbarum plantations in the Ningxia region, mainly in Zhongning County. Goji cultivation also occurs in Gansu, Qinghai Province, and the Haixi Mongol and Tibetan Autonomous Prefecture. The main berry products for export are manufactured beverages, juice concentrate, dried berries, snacks, and freeze-dried juice powder and berries. Ningxia goji has been cultivated along the fertile floodplains of the Yellow River over centuries. The region has developed an industrial association of growers, processors, marketers, and scholars of wolfberry cultivation to promote the berry's commercial and export potential. Ningxia goji berries, the variety used by practitioners of traditional Chinese medicine, are celebrated annually with a festival.
=== Diet === Diet is a critical component of treatment and is in some cases effective on its own following treatment with insulin. For example, a recent mini-study showed that many diabetic cats stopped needing insulin after changing to a low carbohydrate diet. The rationale is that a low-carbohydrate diet reduces the amount of insulin needed and keeps the variation in blood sugar low and easier to predict. Additionally, research indicates that feeding cats low-carbohydrate meals increases their chances of obtaining diabetic remission by four times. Also, fats and proteins are metabolized slower than carbohydrates, reducing dangerous blood sugar peaks right after meals. Cats are obligate carnivores and must consume meat to survive, as their bodies are unable to produce certain amino acids such as taurine and arginine, and cannot process many of the nutrients that meat has from other dietary sources. It is recommended to see a veterinary professional to discuss the specific dietary needs a cat with diabetes needs to manage their diabetes while also ensuring they do not develop other health issues related to malnutrition.

With its central position in Europe, Germany is a transport hub for the continent. Its road network is among the densest in Europe. The motorway (Autobahn) is widely known for having no general federally mandated speed limit for some classes of vehicles. The Intercity Express or ICE train network serves major German cities as well as destinations in neighbouring countries with speeds up to 300 km/h (190 mph). The largest German airports are Frankfurt Airport, Munich Airport and Berlin Brandenburg Airport. The Port of Hamburg is the third-busiest port in Europe and one of the twenty largest container ports in the world. In 2019, Germany was the world's seventh-largest consumer of energy. All German nuclear power plants were phased out in 2023. Germany meets its power demands using 40% renewable sources (2018), and has been called an "early leader" in solar panels and offshore wind. The German energy transition (Energiewende) is the recognised move to a sustainable economy by means of energy efficiency and renewable energy, with the country being called "the world's first major renewable energy economy". Germany has reduced its primary energy consumption by 11% between 1990 and 2015 and set itself goals of reducing it by 30% until 2030 and 50% by 2050. The country is committed to the Paris Agreement and several other treaties promoting biodiversity, low emission standards, and water management. As of 2017, Germany's household recycling rate is among the highest in the world, at around 65%. In 2023, Germany was the 14th highest emitting nation of greenhouse gases.
=== Skin === Flucloxacillin is used for both staphylococcal and streptococcal skin infections. These include folliculitis, carbuncles, impetigo, ecthyma, cellulitis, erysipelas, necrotising fasciitis, and infections of skin conditions such as eczema, scabies, ulcers and acne. Due to the widespread belief that dual-therapy is needed to cover both Staphylococcus and Streptococcus in cellulitis, flucloxacillin is sometimes given with the addition of benzylpenicillin for more severe cellulitis. However, support for this practice has lessened since findings in a study published in the Emergency Medicine Journal in 2005 did not show this combination to give additional clinical benefit. In the UK, using flucloxacillin alone is the first choice for treating cellulitis; other countries vary in their practice.

=== Automation === In the past, oligonucleotide synthesis was carried out manually in solution or on solid phase. The solid phase synthesis was implemented using, as containers for the solid phase, miniature glass columns similar in their shape to low-pressure chromatography columns or syringes equipped with porous filters. Currently, solid-phase oligonucleotide synthesis is carried out automatically using computer-controlled instruments (oligonucleotide synthesizers) and is technically implemented in column, multi-well plate, and array formats. The column format is best suited for research and large scale applications where a high-throughput is not required. Multi-well plate format is designed specifically for high-throughput synthesis on small scale to satisfy the growing demand of industry and academia for synthetic oligonucleotides.
=== Pain === Methadone is used as an analgesic in chronic pain, often in rotation with other opioids. Due to its activity at the NMDA receptor, it may be more effective against neuropathic pain; for the same reason, tolerance to the analgesic effects may be less than that of other opioids.
Varicella vaccine, also known as chickenpox vaccine, is a vaccine that protects against chickenpox. One dose of vaccine prevents 95% of moderate disease and 100% of severe disease. Two doses of vaccine are more effective than one. If given to those who are not immune within five days of exposure to chickenpox it prevents most cases of the disease. Vaccinating a large portion of the population also protects those who are not vaccinated. It is given by injection just under the skin. Another vaccine, known as zoster vaccine, is used to prevent diseases caused by the same virus – the varicella zoster virus. The World Health Organization (WHO) recommends routine vaccination only if a country can keep more than 80% of people vaccinated. If only 20% to 80% of people are vaccinated it is possible that more people will get the disease at an older age and outcomes overall may worsen. Either one or two doses of the vaccine are recommended. In the United States two doses are recommended starting at twelve to fifteen months of age. As of 2017, twenty-three countries recommend all non-medically exempt children receive the vaccine, nine recommend it only for high-risk groups, three additional countries recommend use in only parts of the country, while other countries make no recommendation. Not all countries provide the vaccine due to its cost. In the United Kingdom, the varicella vaccine has been added in 2026 to the routine children vaccination, combined with the MMR vaccine, at ages 12 and 18 months. Minor side effects may include pain at the site of injection, fever, and rash.
== Freeze-dried sculptures == In an attempt to solve a holographic problem, Gibson experimented with freeze-drying techniques. He produced a series of sculptures that explored the ethics of using legally embalmed animals and humans as art supplies. These sculptures were first exhibited at the Unit/Pitt Gallery in Vancouver, Canada in 1981. The same works were later shown in November 1984 at the Cuts Gallery in London, UK.

It is a synthetic peptide of 39 amino acids bearing a lipid side chain. Its size and architecture place it outside the small-molecule class, and laboratories generally handle it with the precautions used for biologic-like molecules.
The fatty diacid side chain promotes strong binding to serum albumin, which slows clearance and yields a half-life of roughly five days. That profile supports once-weekly administration in clinical use.
In vitro assays detect activity at both the GIP and GLP-1 receptors, but the activity ratio depends on the assay system and the signalling pathway measured. The relative contribution of each receptor to clinical effects is still being characterised.
Water promotes hydrolysis and deamidation, so removing it slows degradation during transport and storage. The dry solid is also less prone to microbial growth than a solution. Reconstitution is therefore performed close to the point of use.
