Degradation pathways for tirzepatide include deamidation, oxidation, and aggregation, which are common for therapeutic peptides. These processes can be monitored by size-exclusion chromatography (SEC) for aggregates and ion-exchange chromatography for charge variants. Forced degradation studies under acidic, basic, oxidative, and thermal stress help identify potential impurities. The exact stability profile depends on formulation, concentration, and container-closure system.
Analytical characterization of tirzepatide typically employs reversed-phase high-performance liquid chromatography (RP-HPLC) for purity assessment and peptide mapping. Mass spectrometry, often coupled with electrospray ionization, confirms molecular weight and sequence integrity. Amino acid analysis and capillary electrophoresis may also be used to detect impurities or degradation products. These methods are essential for batch release and stability studies.
Storage recommendations for tirzepatide generally specify refrigeration at 2–8 °C to maintain stability. The peptide should be protected from light and kept in its original packaging to prevent aggregation or adsorption. Freezing is not recommended because freeze-thaw cycles can cause aggregation or precipitation. Once dispensed, storage conditions and in-use periods follow product-specific labeling, which may allow room temperature storage for a limited time.
Cold-chain handling is standard for formulated product, with dry powder stored frozen and ready-to-use solutions refrigerated. Light exposure is minimized because photodegradation of certain amino acid side chains is possible. Shipping and temperature-excursion studies are used to establish whether short deviations affect quality attributes. Documentation supplied with research material usually includes a certificate of analysis listing purity, identity confirmation, and water or residual solvent content. Users are expected to confirm that material meets the stated specification before use.
Identity and purity of tirzepatide are assessed mainly by reversed-phase high-performance liquid chromatography with ultraviolet detection, often paired with mass spectrometry. Because the molecule carries several modifications, gradient conditions are adjusted to resolve the intact peptide from deamidation and oxidation products. Enzymatic digestion followed by peptide mapping confirms the primary sequence and locates specific modifications. Quantitation in biological matrices typically uses liquid chromatography with tandem mass spectrometry after solid-phase extraction. Immunoassays are used less often, since antibody cross-reactivity with closely related peptides can bias results.
The peptide shares degradation routes common to modified peptides: deamidation of asparagine and glutamine residues, oxidation of methionine, and backbone hydrolysis under extreme pH. Lyophilized material is generally more stable than a solution, and residual water content directly affects the rate of hydrolysis. In liquid form, aggregation and visible particles can appear after agitation or repeated freeze-thaw cycles. Stability studies therefore track monomer content, aggregate content, and potency over months under defined temperature and humidity.

Development of tirzepatide took place under a research program that sought to test whether simultaneous engagement of two incretin receptors would produce greater metabolic effects than single-receptor agonism. Clinical trials were organized into the SURPASS series for type 2 diabetes and the SURMOUNT series for obesity and weight management. Regulatory clearance for type 2 diabetes came in 2022 in the United States, followed by approval for chronic weight management in 2023. The trial programs reported reductions in glycated hemoglobin and body weight relative to comparators, though long-term cardiovascular and durability data continue to accumulate.
The peptide backbone contains 39 amino acids and includes alpha-aminoisobutyric acid residues, which are not among the standard proteinogenic set. A C20 fatty diacid moiety is attached through a linker, allowing the compound to bind serum albumin and extend its circulation time. This albumin binding is the main reason the molecule supports once-weekly administration rather than more frequent dosing. The measured molecular mass is approximately 4,813 daltons, placing it firmly in the peptide rather than small-molecule class.
Tirzepatide is a synthetic peptide that activates both the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. This dual agonist profile distinguishes it from earlier incretin-based compounds that act on a single receptor. The molecule was engineered from the native GIP sequence and carries several non-natural residues that slow enzymatic breakdown. Researchers designed it to combine the insulinotropic effects of GIP signaling with the appetite and gastric-emptying effects associated with GLP-1 activation.
Tirzepatide is a synthetic peptide built from 39 amino acid residues. Its sequence is related to human glucose-dependent insulinotropic polypeptide, with modifications that include a C-terminal extension and a C20 fatty diacid joined through a linker. Those changes raise the molecule's affinity for serum albumin, which slows renal filtration and lengthens the time it stays in circulation. The free base has an average molecular mass near 4813.5 daltons. The compound is made by solid-phase peptide synthesis followed by chromatographic purification.
At the receptor level, tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. Both belong to the class B family of G protein-coupled receptors and signal largely through cyclic AMP accumulation. The compound binds the two receptors with differing affinity, and the pattern of signaling at each site is described in the literature as biased rather than simply proportional to occupancy. Tissues carrying these receptors include pancreatic islets, adipose tissue, the central nervous system, and the gastrointestinal tract. The relative weight of each receptor population in producing metabolic effects continues to be studied.
The compound first appeared in the scientific literature as an investigational agent for type 2 diabetes. Clinical development proceeded through phase 1, phase 2, and phase 3 programs that measured glycemic control as a primary endpoint while recording body weight as a secondary outcome. Regulatory approval in the United States followed in 2022 for glycemic control, and a separate indication for chronic weight management was added later. Subsequent trials have examined cardiovascular outcomes in adults with elevated cardiovascular risk. Debates continue over how much of the observed effect derives from each receptor arm.
Structural work on the molecule centers on a C20 fatty diacid moiety attached through a linker to the peptide backbone. This side chain promotes reversible binding to serum albumin, which slows renal clearance and supports a prolonged action profile. The peptide backbone incorporates aminoisobutyric acid substitutions that limit recognition by digestive enzymes. Together these modifications produce a molecule that is stable enough for subcutaneous delivery but still dependent on careful manufacturing control. Analytical characterization of the active pharmaceutical ingredient typically follows the conventions used for other synthetic peptides.
Tirzepatide is a synthetic peptide composed of 39 amino acids. It acts as a dual agonist at two incretin receptors, the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. The molecule was designed by modifying the native sequence of glucose-dependent insulinotropic polypeptide to improve metabolic stability and extend its circulation time. Its structure includes several non-natural amino acid residues and a fatty acid side chain. These features distinguish it from earlier single-receptor incretin analogs studied in the same period.
An extended fatty diacid moiety promotes binding to serum albumin, which slows renal clearance and extends the circulating half-life to roughly five days. That property supports once-weekly administration and largely explains the dosing interval described in clinical reports. Published data come mainly from large randomised programmes that evaluated glycaemic control and body weight over periods of many months. Long-term outcomes beyond those trial windows, including what happens after treatment stops, remain an active area of investigation.
Tirzepatide is a synthetic peptide built from thirty-nine amino acids. Its sequence is derived from native glucose-dependent insulinotropic polypeptide, or GIP, with several non-natural residues and a fatty diacid side chain attached through a linker. The molecule behaves as a dual agonist at two incretin receptors, GIP and GLP-1, instead of targeting a single receptor. This dual engagement separates it from earlier single-receptor incretin compounds and underpins most of its reported pharmacological activity.
At the receptor level, the compound binds both GIP and GLP-1 receptors and triggers downstream signalling that raises cyclic AMP in target cells. GLP-1 receptor activation is associated with glucose-dependent insulin release, slower gastric emptying, and reduced appetite signalling. GIP receptor activation contributes effects that are less completely characterised, and how much each receptor adds to the overall clinical response is still an open question. The two pathways appear to interact in a complementary rather than a purely additive way.
==== 8 April ==== In Iran, the Lavan Oil Refinery, located on Lavan Island was struck at around 10 a.m.; a fire ignited. The Iranian authorities denounced the event and stated that it was an act of "cowardice" by the "enemies". Israel denied being involved in the attack. The debris of a drone shot down by Iranian forces killed a 7-year-old child and injured another six members of the child's family in the town of Shushtar in Khuzestan province.
Going hitless in 16 at bats during the World Series that fall, in addition to his feud with manager Dick Williams over lack of playing time, resulted in the Athletics fulfilling his trade demand by sending him to the Texas Rangers for Horacio Piña on December 1, 1972. Additionally, the A's wanted to free up the first base position for Gene Tenace who was the star of that same Fall Classic. Opening 1973 batting .188 with one homer and six RBIs, he was dealt along with Rich Hand and Rick Stelmaszek from the Rangers to the California Angels for Jim Spencer and Lloyd Allen on May 20. In 1973, he was seventh in the league in hit by pitches (8). On May 4, 1974, he was released by the Angels. In 907 games over nine seasons, Epstein posted a .244 batting average (695-for-2,854) with 362 runs, 130 home runs, 380 RBIs, 448 bases on balls, .358 on-base percentage and .424 slugging percentage. He finished his career with a .991 fielding percentage playing every inning at first base. In 13 postseason games, he hit only .108 (4-for-37) with two runs scored, one home run, one RBI, and nine walks. In 1991 he was inducted into the Southern California Jewish Sports Hall of Fame. He was inducted as a member of the United States National Jewish Sports Hall of Fame in 2004. Through 2010, he was sixth all-time in career home runs (behind Mike Lieberthal) among Jewish major league baseball players.
In those with stable coronary artery disease, but no heart failure, benefits are similar to other usual treatments. In 2012, a meta-analysis published in the BMJ described the protective role of ACE inhibitors in reducing the risk of pneumonia when compared to angiotensin II receptor blocker (ARBs). The authors found a decreased risk in patients with previous stroke (54% risk reduction), with heart failure (37% risk reduction), and of Asian descent (43% risk reduction vs 54% risk reduction in non-Asian population). However, no reduced pneumonia-related mortality was observed. ACE inhibitors have been shown to be effective for indications other than hypertension even in patients with normal blood pressure. The use of a maximum dose of ACE inhibitors in such patients (including for prevention of diabetic nephropathy, congestive heart failure, and prophylaxis of cardiovascular events) is justified, because it improves clinical outcomes independently of the blood pressure-lowering effect of ACE inhibitors. Such therapy, of course, requires careful and gradual titration of the dose to prevent the effects of rapidly decreasing blood pressure (dizziness, fainting, etc.). ACE inhibitors may also be used to help decrease excessive water consumption in people with schizophrenia resulting in psychogenic polydipsia. A double-blind, placebo-controlled trial showed that when used for this purpose, enalapril led to decreased consumption (determined by urine output and osmolality) in 60% of people; the same effect has been demonstrated in other ACE inhibitors.
If a female rat is given testosterone in the first few days of postnatal life, during the "critical period" of sex-steroid influence in rats, the hypothalamus is irreversibly defeminized and masculinized; the adult rat will be incapable of generating an LH surge in response to estrogen as is characteristic of females, but will be capable of exhibiting male sexual behaviors e.g. mounting a sexually receptive female. By contrast, a male rat castrated just after birth will be feminized, and the adult will show typical female "receptive" sexual behavior in response to estrogen, that is, lordosis behavior. Masculinization and feminization can be distinguished from their complimentary de-feminization and de-masculinization, as neonatal treatment with COX2 inhibitors or PgE2 makes it possible to create rats which exhibit neither sexual behaviour, or both, respectively. Some effects of combined masculinization and feminization on hypothalamic physiology are known, but outcomes where the processes oppose (e.g. proportions of cell types) remain unreported in vitro as of 2025. In primates, the developmental influence of androgens is less clear, and the consequences are less understood. Within the brain, testosterone is aromatized (to estradiol), which is the principal active hormone for developmental influences. The human testis secretes high levels of testosterone from about week eight of fetal life until five to six months after birth (a similar perinatal surge in testosterone is observed in many species), a process that appears to underlie the male phenotype.
1RAR trained in Kenya from December 1943 to September 1944, when it transferred to Ceylon and became part of the 22nd (East Africa) Infantry Brigade alongside the 1st KAR and the 3rd Northern Rhodesia Regiment. In December 1944, after three months' training for jungle warfare, 1RAR and the other two components of the brigade joined the Burma Campaign at Chittagong under the command of the 15th Indian Corps. The brigade spent about three months supporting the 25th Indian Division in north-western Burma, advancing through the Mayu peninsula during January 1945 and taking part in the latter stages of the Battle of Ramree Island, landing on the island on 14 February. 1RAR fortified positions at Myinbin, Kyaukkale and Mayin but did not contact Japanese forces. A widespread belief developed among Japanese troops in Burma that the British Army's African soldiers were cannibals, partly because of deliberate disinformation spread by the black troops themselves as they travelled around the country. While entirely unfounded, the notion "that we Africans eat people", as one RAR soldier put it, had a fearsome psychological effect; men of 1RAR reported Japanese soldiers picking up their comrades' bodies in the midst of battle and running away. In March 1945 the 22nd Brigade was ordered south to Dalaba where it became part of the 82nd (West Africa) Division, which had been tasked with clearing the Taungup area of Japanese troops.

During atrial systole, blood flows from the atria to the ventricles down the pressure gradient. Chordae tendineae are relaxed because the atrioventricular (AV) valves are forced open. When the ventricles of the heart contract in ventricular systole, the increased blood pressures in both chambers push the AV valves to close simultaneously, preventing the backflow of blood into the atria. Since the blood pressure in the atria is much lower than that in the ventricles, the flaps attempt to evert to the low pressure regions. The chordae tendineae prevent this prolapse by becoming tense, which pulls on the flaps, holding them in a closed position.
In July 2020, the European Medicines Agency (EMA) started reviewing results from the RECOVERY study arm that involved the use of dexamethasone in the treatment of patients with COVID-19 admitted to the hospital to provide an opinion on the results and in particular the potential use of dexamethasone for the treatment of adults with COVID-19. In September 2020, the EMA received an application for marketing authorization of dexamethasone for COVID-19.

=== Precursor to other peroxide compounds === Hydrogen peroxide is a weak acid, forming hydroperoxide or peroxide salts with many metals. It also converts metal oxides into the corresponding peroxides. For example, upon treatment with hydrogen peroxide, chromic acid (CrO3 and H2SO4) forms a blue peroxide CrO(O2)2.

Arias was elected president in 1968 following a populist campaign. Soon after taking office he launched a purge of the National Guard, sending much of its general staff into "diplomatic exile" or retirement. In response, Torrijos and a few other officers led a coup against him, ousting him after an eleven-day presidency. The coup was set in motion by Martínez, as the leader of the garrison at Chiriquí, and received the support of most military officers. A power struggle followed between the various forces involved in the coup, and chiefly between Torrijos and Martínez. Noriega was an important supporter of Torrijos during this conflict. In February 1969, Torrijos's men seized Martínez and exiled him to Miami giving Torrijos control of the country. At the end of 1969, Torrijos went to Mexico on holiday. A coup was launched in his absence, in which Noriega's loyalty allowed Torrijos to hang on to power, greatly enhancing Torrijos's image. Noriega was promoted to captain a month after the coup attempt: just 18 months later, in August 1970, Torrijos promoted him to the position of lieutenant colonel and appointed him chief of military intelligence. According to Dinges, by this point Noriega had left his undisciplined past behind him. When Arias's supporters launched a guerrilla uprising in his home province, Noriega as the head of intelligence played an important role in putting it down within a year. Torrijos retained power as a military ruler until 1981: during this time he negotiated the Torrijos–Carter Treaties with U.S.
