The short version of dual agonist fits in a sentence. The long version — which is the one that helps — is below.

Dual agonism at the GIP and GLP-1 receptors underlies the observed pharmacology. Activation of GLP-1 receptors raises glucose-dependent insulin release, lowers glucagon secretion, slows gastric emptying and reduces appetite. GIP receptor activation contributes additional effects on adipose tissue and on energy balance, and the combined action on appetite appears larger than either pathway alone in animal models. Signalling bias and the relative contribution of each receptor arm to weight-related effects remain areas of active investigation.
Structure-activity work shows that fatty acid length, linker chemistry and the position of acylation all influence albumin affinity and receptor potency. Plasma protein binding exceeds 99 percent, which restricts distribution and slows renal clearance. Degradation proceeds largely through general proteolysis and fatty acid oxidation rather than cytochrome P450 metabolism, so exposure to common oxidative drug interactions is limited. Whether these clearance routes vary meaningfully between individuals is not fully established.
The molecule is a synthetic 39-amino-acid peptide whose backbone derives from the sequence of human glucose-dependent insulinotropic polypeptide, with several substitutions that raise metabolic stability and shift receptor preference. A C20 fatty diacid is attached through a short linker to a lysine side chain, a modification that increases binding to serum albumin. The reported monoisotopic mass is approximately 4813 Da. Near neutral pH the peptide carries a net negative charge, and the lipid tail makes the molecule markedly more hydrophobic than the unmodified parent sequence.
Published work supports the view that engaging two incretin receptors produces changes in glucose handling and body weight larger than those seen with single-receptor activation. Why that difference arises is not fully settled. Open questions include how much of the observed weight effect depends on central versus peripheral signaling, and whether the two receptors form interacting complexes. Most reported findings come from controlled trials and animal models, and translation between species is imperfect. Further research is expected to refine these points over time.
Tirzepatide is a synthetic peptide built from 39 amino acid residues. Its sequence is related to human glucose-dependent insulinotropic polypeptide, with modifications that include a C-terminal extension and a C20 fatty diacid joined through a linker. Those changes raise the molecule's affinity for serum albumin, which slows renal filtration and lengthens the time it stays in circulation. The free base has an average molecular mass near 4813.5 daltons. The compound is made by solid-phase peptide synthesis followed by chromatographic purification.
At the receptor level, tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. Both belong to the class B family of G protein-coupled receptors and signal largely through cyclic AMP accumulation. The compound binds the two receptors with differing affinity, and the pattern of signaling at each site is described in the literature as biased rather than simply proportional to occupancy. Tissues carrying these receptors include pancreatic islets, adipose tissue, the central nervous system, and the gastrointestinal tract. The relative weight of each receptor population in producing metabolic effects continues to be studied.

Recommended storage for reference material is a freezer at approximately -20 degrees Celsius, protected from light and moisture. Commercial injectable presentations are stored refrigerated between 2 and 8 degrees Celsius and must not be frozen. Product labelling generally permits a limited period at controlled room temperature once dispensed, with the exact window depending on the presentation. Repeated temperature cycling is avoided because it can promote aggregation or deamidation of the peptide chain.
Identity and purity are assessed by reversed-phase high-performance liquid chromatography, with mass confirmation by electrospray ionisation mass spectrometry. Peptide mapping after enzymatic digestion verifies the primary sequence. Size-exclusion chromatography quantifies aggregates, while circular dichroism provides a secondary-structure fingerprint. Bioanalytical quantification in plasma uses immunoassay or LC-MS/MS. Reported purity for research-grade lots is commonly 95 percent or higher, and residual water content is checked by Karl Fischer titration.
Reported outcomes in large trials include dose-dependent weight reduction and improvements in glycemic markers over periods ranging from several months to more than a year. Whether the compound alters long-term cardiovascular or renal outcomes is being examined in dedicated outcome studies, so those questions remain open. Labeling describes gastrointestinal effects such as nausea and diarrhea, which tend to appear during dose escalation. Discontinuation rates and the durability of effects after treatment stops vary across study populations and are still debated.
Tirzepatide is a synthetic peptide developed as a dual agonist at the glucose-dependent insulinotropic polypeptide and glucagon-like peptide-1 receptors. Its structure is built on a GIP-derived backbone with non-natural amino acid substitutions and a fatty diacid side chain that promotes albumin binding and slows clearance. That modification supports once-weekly subcutaneous dosing. Registrational trial programs reported reductions in body weight and glycated hemoglobin alongside the drug's glycemic effects.
Clinical research programs have evaluated tirzepatide in adults with type 2 diabetes and in adults with obesity or excess weight. Trials generally reported reductions in glycated hemoglobin and body weight across treatment periods of several months. Since these studies enrolled defined populations under controlled conditions, the findings describe group averages rather than individual outcomes. Open questions include the durability of effects after treatment stops, variation among subgroups, and the long-term consequences of sustained dual receptor stimulation. Published trial summaries should be consulted for exact measurements rather than secondary accounts.
Tirzepatide is a synthetic peptide built from 39 amino acid residues. Its backbone derives from the native glucose-dependent insulinotropic polypeptide sequence, altered at several positions to resist enzymatic cleavage. A fatty diacid group attached through a linker extends plasma residence time by promoting reversible binding to serum albumin. The molecule carries a net negative charge near physiological pH and has a reported molecular weight close to 4813 daltons. These features separate it from shorter incretin analogs and account for its prolonged dosing interval.
People with diabetes can benefit from education about the disease and treatment, dietary changes, and exercise, with the goal of keeping both short-term and long-term blood glucose levels within acceptable bounds. In addition, given the associated higher risks of cardiovascular disease, lifestyle modifications are recommended to control blood pressure. Weight loss can prevent progression from prediabetes to diabetes type 2, decrease the risk of cardiovascular disease, or result in a partial remission in people with diabetes. No single dietary pattern is best for all people with diabetes. Healthy dietary patterns, such as the Mediterranean diet, low-carbohydrate diet, or DASH diet, are often recommended, although evidence does not support one over the others. According to the ADA, "reducing overall carbohydrate intake for individuals with diabetes has demonstrated the most evidence for improving glycemia", and for individuals with type 2 diabetes who cannot meet the glycemic targets or where reducing anti-glycemic medications is a priority, low or very-low carbohydrate diets are a viable approach. For overweight people with type 2 diabetes, any diet that achieves weight loss is effective. A 2020 Cochrane systematic review compared several non-nutritive sweeteners to sugar, placebo and a nutritive low-calorie sweetener (tagatose), but the results were unclear for effects on HbA1C, body weight and adverse events.

Hyperinsulinism has also been implicated as a contributing factor in the excessive production of androgens in polycystic ovary syndrome. The principal treatments of hyperinsulinism due to insulin resistance are measures that improve insulin sensitivity, such as weight loss, physical exercise, and drugs such as thiazolidinediones or metformin.
In an exothermic reaction, the temperature in the sample cell increases upon addition of ligand. This causes the feedback power to the sample cell to be decreased (remember: a reference power is applied to the reference cell) in order to maintain an equal temperature between the two cells. In an endothermic reaction, the opposite occurs; the feedback circuit increases the power in order to maintain a constant temperature (isothermal operation). Observations are plotted as the power needed to maintain the reference and the sample cell at an identical temperature against time. As a result, the experimental raw data consists of a series of spikes of heat flow (power), with every spike corresponding to one ligand injection. These heat flow spikes/pulses are integrated with respect to time, giving the total heat exchanged per injection. By integrating each peak from the baseline, the total heat associated with each injection is obtained, including both reaction-specific and non-reaction-related contributions. The pattern of these heat effects as a function of the molar ratio [ligand]/[macromolecule] can then be analyzed to give the thermodynamic parameters of the interaction under study.
Feathers are a feature characteristic of birds (though also present in some dinosaurs not currently considered to be true birds). They facilitate flight, provide insulation that aids in thermoregulation, and are used in display, camouflage, and signalling. There are several types of feathers, each serving its own set of purposes. Feathers are epidermal growths attached to the skin and arise only in specific tracts of skin called pterylae. The distribution pattern of these feather tracts (pterylosis) is used in taxonomy and systematics. The arrangement and appearance of feathers on the body, called plumage, may vary within species by age, social status, and sex. Plumage is regularly moulted; the standard plumage of a bird that has moulted after breeding is known as the "non-breeding" plumage, or—in the Humphrey–Parkes terminology—"basic" plumage; breeding plumages or variations of the basic plumage are known under the Humphrey–Parkes system as "alternate" plumages. Moulting is annual in most species, although some may have two moults a year, and large birds of prey may moult only once every few years. Moulting patterns vary across species. In passerines, flight feathers are replaced one at a time with the innermost primary being the first. When the fifth of sixth primary is replaced, the outermost tertiaries begin to drop. After the innermost tertiaries are moulted, the secondaries starting from the innermost begin to drop and this proceeds to the outer feathers (centrifugal moult). The greater primary coverts are moulted in synchrony with the primary that they overlap.

National Security Agency monitored a telephone conversation between Noriega and Luis Córdoba, the military commander in Chiriquí province where Spadafora was arrested. During the conversation Córdoba told Noriega, "We have the rabid dog." Noriega responded, "And what does one do with a dog that has rabies?" Spadafora's murder badly damaged Noriega's image, both within and outside Panama, and created a crisis for the Panamanian regime. Barletta, who was in New York City when Spadafora was murdered in September 1985, announced his intention to appoint an independent commission to investigate the murder. Upon his return to Panama, however, he was forced to resign by the PDF and was replaced by Vice President Eric Arturo Delvalle. Barletta was highly regarded in the Reagan administration, and his removal brought a downturn in the relations between the U.S. and Noriega. After Spadafora's murder the U.S. began to view Noriega as a liability rather than an asset, despite his ongoing support for U.S. interventions elsewhere. The U.S. response included reducing economic assistance and pressuring Panama to reform its banking secrecy laws, crack down on narcotics trafficking, investigate the murder of Spadafora, and reduce the PDF's role in the government. The response to Spadafora's murder created divisions within the PRD, and further damaged the credibility of the government-controlled news media.
Massachusetts stating that crime laboratory reports may not be used against criminal defendants at trial unless the analysts responsible for creating them give testimony and subject themselves to cross-examination. The Supreme Court cited the National Academies of Sciences report Strengthening Forensic Science in the United States in their decision. Writing for the majority, Justice Antonin Scalia referred to the National Research Council report in his assertion that "Forensic evidence is not uniquely immune from the risk of manipulation." In the US, another area of forensic science that has come under question in recent years is the lack of laws requiring the accreditation of forensic labs. Some states require accreditation, but some states do not. Because of this, many labs have been caught performing very poor work resulting in false convictions or acquittals. For example, it was discovered after an audit of the Houston Police Department in 2002 that the lab had fabricated evidence which led George Rodriguez being convicted of raping a fourteen-year-old girl. The former director of the lab, when asked, said that the total number of cases that could have been contaminated by improper work could be in the range of 5,000 to 10,000. The Innocence Project database of DNA exonerations shows that many wrongful convictions contained forensic science errors. According to the Innocence project and the US Department of Justice, forensic science has contributed to about 39 percent to 46 percent of wrongful convictions.
Valve Corporation, also known as Valve Software, is an American video game developer, publisher, hardware, and digital distribution company headquartered in Bellevue, Washington. It is the developer of game franchises including Half-Life, Counter-Strike, Portal, Team Fortress, Left 4 Dead and Dota, the software service Steam, and hardware including the Steam Frame, Steam Machine, Steam Deck and Valve Index. Valve was founded in 1996 by former Microsoft employees Gabe Newell and Mike Harrington. Their debut game, the first-person shooter (FPS) Half-Life (1998), was a critical and commercial success that had a lasting influence on the genre. Harrington left in 2000. In 2003, Valve launched Steam, followed by Half-Life 2 (2004), the episodic sequels Half-Life 2: Episode One (2006) and Episode Two (2007), the puzzle games Portal (2007) and Portal 2 (2011), and the multiplayer games Team Fortress 2 (2007), Left 4 Dead (2008), and Dota 2 (2013). During the 2010s, Valve released fewer games and focused on hardware and virtual reality (VR). They released the first Steam Machine console in 2015, the Valve Index VR headset in 2019, their flagship VR game, Half-Life: Alyx, in 2020, and the handheld system Steam Deck in 2022. In 2026, Valve released a redesigned Steam Machine and a standalone VR headset, the Steam Frame. Valve uses a flat organizational structure, allowing employees to choose their own projects. Valve is noted for its technical and artistic innovation, with many of its games receiving mention among the greatest games of all time.
Adolescents experiencing gender dysphoria may opt to undergo puberty-suppressing hormone therapy at the onset of puberty. The Standards of Care set forth by WPATH recommend individuals pursuing puberty-suppressing hormone therapy wait until at least experiencing Tanner Stage 2 pubertal development. Tanner Stage 2 is defined by the appearance of scant pubic hair, breast bud development, and/or slight testicular growth. WPATH classifies puberty-suppressing hormone therapy as a "fully reversible" intervention. Delaying puberty allows individuals more time to explore their gender identity before deciding on more permanent interventions and prevents the physical changes associated with puberty. The preferred puberty-suppressing agent for both individuals assigned male at birth and individuals assigned female at birth is a GnRH Analogue. This approach temporarily shuts down the Hypothalamic-Pituitary-Gonadal (HPG) Axis, which is responsible for the production of hormones (estrogen, testosterone) that cause the development of secondary sexual characteristics in puberty. According to a study by JAMA Pediatrics published in January 2025, less than 0.1% of adolescents covered by private medical insurance in the US take gender-affirming medication to treat gender dysphoria.

=== Catalyst === In 2014, researchers at the University of Western Australia discovered nano sized fragments of graphene can speed up the rate of chemical reactions. In 2015, researchers announced an atomic scale catalyst made of graphene doped with nitrogen and augmented with small amounts of cobalt whose onset voltage was comparable to platinum catalysts. In 2016 iron-nitrogen complexes embedded in graphene were reported as another form of catalyst. The new material was claimed to approach the efficiency of platinum catalysts. The approach eliminated the need for less efficient iron nanoparticles.
Because of the extraordinary coupling efficiency of HATU, it has often been used for intramolecular amidation (coupling of a carboxylic acid and an amine of the same molecule). For example, the formation of cyclo-tetrapeptides through the head-to-tail reaction of linear tetrapeptides assisted by HATU has been reported.
=== Right lung === The right lung has both more lobes and segments than the left. It is divided into three lobes, an upper, middle, and a lower lobe by two fissures, one oblique and one horizontal. The upper, horizontal fissure, separates the upper from the middle lobe. It begins in the lower oblique fissure near the posterior border of the lung, and, running horizontally forward, cuts the anterior border on a level with the sternal end of the fourth costal cartilage; on the mediastinal surface it may be traced back to the hilum. The lower, oblique fissure, separates the lower from the middle and upper lobes and is closely aligned with the oblique fissure in the left lung. The mediastinal surface of the right lung is indented by a number of nearby structures. The heart sits in an impression called the cardiac impression. Above the hilum of the lung is an arched groove for the azygos vein, and above this is a wide groove for the superior vena cava and right brachiocephalic vein; behind this, and close to the top of the lung is a groove for the brachiocephalic artery. There is a groove for the oesophagus behind the hilum and the pulmonary ligament, and near the lower part of the oesophageal groove is a deeper groove for the inferior vena cava before it enters the heart. The weight of the right lung varies between individuals, with a standard reference range in men of 155–720 g (0.342–1.587 lb) and in women of 100–590 g (0.22–1.30 lb).
In the United States, Utah started screening for GAMT deficiency in all newborns in 2015. New York started screening newborns in late 2018. In 2020, a GAMT-positive infant was identified via newborn screening in Utah, thus providing evidence that a case could be identified from newborn screening. A second infant was identified in New York in 2021. In 2022, a federal advisory committee voted to include GAMT in the Recommended Universal Screening Panel starting in January 2023. In addition to New York and Utah, California, Pennsylvania, Connecticut, Delaware, and Michigan all currently screen for GAMT. Around the world, the state of Victoria, Australia began screening for GAMT in 2004, leading to the diagnosis of a newborn at birth in 2022. In Canada, the provinces of Ontario and British Columbia screen for GAMT.
The unusual stability of the helium-4 nucleus is also important cosmologically: it explains the fact that in the first few minutes after the Big Bang, as the "soup" of free protons and neutrons which had initially been created in about 6:1 ratio cooled to the point that nuclear binding was possible, almost all first compound atomic nuclei to form were helium-4 nuclei. Owing to the relatively tight binding of helium-4 nuclei, its production consumed nearly all of the free neutrons in a few minutes, before they could beta-decay, and thus few neutrons were available to form heavier atoms such as lithium, beryllium, or boron. Helium-4 nuclear binding per nucleon is stronger than in any of these elements (see nucleogenesis and binding energy) and thus, once helium had been formed, no energetic drive was available to make elements 3, 4 and 5. It is barely energetically favorable for helium to fuse into the next element with a lower energy per nucleon, carbon. However, due to the short lifetime of the intermediate beryllium-8, this process requires three helium nuclei striking each other nearly simultaneously (see triple-alpha process). There was thus no time for significant carbon to be formed in the few minutes after the Big Bang, before the early expanding universe cooled to the temperature and pressure point where helium fusion to carbon was no longer possible. This left the early universe with a very similar ratio of hydrogen/helium as is observed today (3 parts hydrogen to 1 part helium-4 by mass), with nearly all the neutrons in the universe trapped in helium-4.

It is a synthetic peptide of 39 amino acids bearing a lipid side chain. Its size and architecture place it outside the small-molecule class, and laboratories generally handle it with the precautions used for biologic-like molecules.
The fatty diacid side chain promotes strong binding to serum albumin, which slows clearance and yields a half-life of roughly five days. That profile supports once-weekly administration in clinical use.
In vitro assays detect activity at both the GIP and GLP-1 receptors, but the activity ratio depends on the assay system and the signalling pathway measured. The relative contribution of each receptor to clinical effects is still being characterised.
It is a synthetic peptide and a dual agonist of two incretin receptors. It is not a small molecule, and it is not structurally related to the older single-receptor peptide agonists.
