As a peptide, tirzepatide is handled as a lyophilised solid in research settings and as a preserved solution in finished products. Aqueous solubility is pH dependent and reaches a minimum near the isoelectric point, which lies close to pH 5.4. Stock solutions are typically prepared in neutral or slightly basic buffer to limit precipitation. The solid is hygroscopic and should be equilibrated to room temperature before opening so that condensation does not form on the powder surface.
Recommended storage for reference material is a freezer at approximately -20 degrees Celsius, protected from light and moisture. Commercial injectable presentations are stored refrigerated between 2 and 8 degrees Celsius and must not be frozen. Product labelling generally permits a limited period at controlled room temperature once dispensed, with the exact window depending on the presentation. Repeated temperature cycling is avoided because it can promote aggregation or deamidation of the peptide chain.
Identity and purity are assessed by reversed-phase high-performance liquid chromatography, with mass confirmation by electrospray ionisation mass spectrometry. Peptide mapping after enzymatic digestion verifies the primary sequence. Size-exclusion chromatography quantifies aggregates, while circular dichroism provides a secondary-structure fingerprint. Bioanalytical quantification in plasma uses immunoassay or LC-MS/MS. Reported purity for research-grade lots is commonly 95 percent or higher, and residual water content is checked by Karl Fischer titration.
Tirzepatide is a synthetic peptide built from 39 amino acid residues. Its sequence is related to human glucose-dependent insulinotropic polypeptide, with modifications that include a C-terminal extension and a C20 fatty diacid joined through a linker. Those changes raise the molecule's affinity for serum albumin, which slows renal filtration and lengthens the time it stays in circulation. The free base has an average molecular mass near 4813.5 daltons. The compound is made by solid-phase peptide synthesis followed by chromatographic purification.
At the receptor level, tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. Both belong to the class B family of G protein-coupled receptors and signal largely through cyclic AMP accumulation. The compound binds the two receptors with differing affinity, and the pattern of signaling at each site is described in the literature as biased rather than simply proportional to occupancy. Tissues carrying these receptors include pancreatic islets, adipose tissue, the central nervous system, and the gastrointestinal tract. The relative weight of each receptor population in producing metabolic effects continues to be studied.

Dual agonism at the GIP and GLP-1 receptors underlies the observed pharmacology. Activation of GLP-1 receptors raises glucose-dependent insulin release, lowers glucagon secretion, slows gastric emptying and reduces appetite. GIP receptor activation contributes additional effects on adipose tissue and on energy balance, and the combined action on appetite appears larger than either pathway alone in animal models. Signalling bias and the relative contribution of each receptor arm to weight-related effects remain areas of active investigation.
Structure-activity work shows that fatty acid length, linker chemistry and the position of acylation all influence albumin affinity and receptor potency. Plasma protein binding exceeds 99 percent, which restricts distribution and slows renal clearance. Degradation proceeds largely through general proteolysis and fatty acid oxidation rather than cytochrome P450 metabolism, so exposure to common oxidative drug interactions is limited. Whether these clearance routes vary meaningfully between individuals is not fully established.
Long-term storage of lyophilised peptide powder is generally at minus twenty degrees Celsius or colder, with desiccant and protection from light. Short-term storage at two to eight degrees Celsius is common during active use. In solution, stability depends strongly on pH, concentration, and the presence of preservatives, and hydrolysis or aggregation can develop over weeks. Published stability data specific to this molecule are limited, so recommended conditions for research material are usually extrapolated from general peptide handling practice rather than from a dedicated study.
Bulk peptide material is normally characterised by reversed-phase high-performance liquid chromatography, which separates the target sequence from truncation products and other closely related impurities. Ultraviolet detection near 214 nanometres is common because the peptide backbone absorbs in that region. Mass spectrometry, usually electrospray ionisation coupled to a mass analyser, is used to confirm the molecular mass. Because the molecule carries a lipophilic side chain, gradient methods often need a relatively high organic modifier fraction to elute it within a practical retention window.
Like most synthetic peptides of this size, the material is commonly supplied as a lyophilised powder that appears white to off-white. It dissolves in aqueous buffers and in mixtures of water with a small proportion of organic solvent, though the fatty acid portion reduces solubility in pure water relative to short peptides. Hygroscopic behaviour is reported for many peptide powders, so weighing is usually performed quickly and under controlled humidity. Working solutions are typically prepared fresh and kept cold.
=== Medical and dental uses === Casein-derived compounds are used in tooth remineralization products to stabilize amorphous calcium phosphate (ACP) and release the ACP onto tooth surfaces, where it can facilitate remineralization. Casein and gluten exclusion diets are sometimes used in alternative medicine for children with autism. The evidence that such diets have any impact on behavior or cognitive and social functioning in autistic children was poor.
==== United States ==== TMA-2 is not an explicitly controlled substance in the United States. However, it is a positional isomer of 3,4,5-trimethoxyamphetamine (TMA), and thus may be considered a Schedule I controlled substance in this country similarly to TMA.
== History == Olaratumab was originally developed by ImClone Systems, which was acquired by Eli Lilly in 2008. A Phase I clinical trial was conducted in Japanese patients in September 2010, followed by a Phase II trial in 133 patients, starting in October 2010. In February 2015, the European Medicines Agency assigned olaratumab orphan drug status for the treatment of soft-tissue sarcoma. The European Commission granted a conditional marketing authorisation, based on the mentioned Phase II study, valid throughout the European Union on 9 November 2016. Previously considered a promising drug, the FDA granted olaratumab fast track designation, breakthrough therapy designation and priority review status. In October 2016, the US FDA issued an accelerated approval notice for use of olaratumab with doxorubicin to treat adults with certain types of soft-tissue sarcoma, based on the same study. A phase III trial completed in 2019, and unfortunately showed no benefit from the addition of olaratumab to doxorubicin. As noted above, these results led to withdrawal of approval in the United States and Europe.

Many affected infants also have distinctive abnormalities of the head and facial (craniofacial) area. Many of the physical features associated with the disorder are congenital. In most cases, premature closure of fibrous joints (sagittal sutures) between certain bones (parietal bones) of the skull may restrict lateral growth of the skull, causing it to appear abnormally long and narrow that is disproportionate to the body size. In addition, the forehead may be abnormally prominent and the face may be triangular shaped with a hypo plastic mid face, pointed chin. Infants with this disorder may also have abnormally flat cheeks and cheekbones, large ears, prominent mouth with widely spread lips, and or underdeveloped upper jaw bones (maxillary hypoplasia). In addition, in some cases, the teeth may be abnormally crowded together, particularly toward the front of the mouth (anterior crowding) and as a result, the upper result, the upper jaw and lower teeth may not meet properly, they might be abnormally crowded together.
Among the actinides, thorium and uranium are the easiest to isolate. Thorium is extracted mostly from monazite: thorium pyrophosphate (ThP2O7) is reacted with nitric acid, and the produced thorium nitrate treated with tributyl phosphate. Rare-earth impurities are separated by increasing the pH in sulfate solution. In another extraction method, monazite is decomposed with a 45% aqueous solution of sodium hydroxide at 140 °C. Mixed metal hydroxides are extracted first, filtered at 80 °C, washed with water and dissolved with concentrated hydrochloric acid. Next, the acidic solution is neutralized with hydroxides to pH = 5.8 that results in precipitation of thorium hydroxide (Th(OH)4) contaminated with ~3% of rare-earth hydroxides; the rest of rare-earth hydroxides remains in solution. Thorium hydroxide is dissolved in an inorganic acid and then purified from the rare earth elements. An efficient method is the dissolution of thorium hydroxide in nitric acid, because the resulting solution can be purified by extraction with organic solvents:
===== Sweden ===== Cannabidiol is classified as a medical product in Sweden. However, in July 2019, Supreme Court of Sweden ruled that CBD oil with any concentration of THC falls under the narcotic control laws.
On 18 September 2026, the United States, Denmark, and Greenland reached an agreement in which the United States would receive the rights to an expanded military footprint in Greenland with greater security control, without, according to the Danish and Greelandic governments, conditionally transferring much military sovereignty of Greenland to the United States. The agreement was signed on 22 September 2026.

Overall, there are various non-selective calcium channel blockers that help alleviate symptoms of hypertension, schizophrenia, cardiac arrhythmia, epilepsy, pain, asthma, bradycardia, angina pectoris and Alzheimer's disease. Although many of the therapeutic compounds' main target is not P-type channels, further research needs to determine if the clinical effects of these compounds are also influenced by the P-type channel blockage.
== Function == This gene, expressed in the pituitary, encodes a receptor for growth-hormone-releasing hormone. Binding of this hormone to the receptor leads to synthesis and release of growth hormone. Mutations in this gene have been associated with isolated growth-hormone deficiency (IGHD), also known as Dwarfism of Sindh, a disorder characterized by short stature. Many alternate transcriptional splice variants encoding different isoforms have been described, but only two have been characterized to date.
Allied Control Council Law No. 25 on the control of scientific research dated 29 April 1946 restricted German scientists to conducting basic research only, and on 11 July the Allied Control Council dissolved the KWS on the insistence of the Americans, who considered that it had been too close to the national socialist regime, and was a threat to world peace. However, the British, who had voted against the dissolution, were more sympathetic, and offered to let the Kaiser Wilhelm Society continue in the British Zone, on one condition: that the name be changed. Hahn and Heisenberg were distraught at this prospect. To them it was an international brand that represented political independence and scientific research of the highest order. Hahn noted that it had been suggested that the name be changed during the Weimar Republic, but the Social Democratic Party of Germany had been persuaded not to. To Hahn, the name represented the good old days of the German Empire, however authoritarian and undemocratic it was, before the hated Weimar Republic. Heisenberg asked Niels Bohr for support, but Bohr recommended that the name be changed. Lise Meitner wrote to Hahn, explaining that:Outside of Germany it is considered so obvious that the tradition from the period of Kaiser Wilhelm has been disastrous and that changing the name of the KWS is desirable, that no one understands the resistance against it.
== Pathophysiology == Multiple pathophysiological changes have been observed in PAH. This includes an imbalance in apoptosis (programmed cell death) and proliferation of endothelial cells, resulting in intimal thickening as well as proliferation and hyperplasia of the smooth muscle cells constituting the muscular layer of the pulmonary arteries. The smooth muscles in the tunica media also extend more distally than normal, encroaching upon the capillary bed. Infiltration of inflammatory cells, proliferation of fibroblasts and disruptions in collagen architecture result in adventitial thickening and remodeling. All of these changes combine to lead to thickening of the pulmonary arteries and arterioles with an associated increase in pulmonary arterial resistance (increased pulmonary artery pressure). Pathogenic and inappropriate platelet activation coupled with endothelial injury leads to formation of micro-thrombi. And PAH also involve the characteristic plexiform lesions which are growths in the walls of the arterioles consisting of dilated blood vessels which communicate with the bronchial artery and vaso vasorum. As pulmonary hypertension persists and worsens the right ventricle undergoes compensatory changes such as concentric hypertrophy of the heart muscle and changes in the microcirculation. However, with prolonged pulmonary hypertension, with the right ventricle pumping against elevated right heart pressures, the hypertrophy becomes maladaptive with microvascular rarefaction, and fibrosis. These changes eventually culminate in right heart failure.
Instead of in the liquid phase, the dehydrochlorination may also be carried out in the gas phase on a zinc chloride impregnated catalyst in a fluidized bed reactor at 350-500 °C. The seemingly simple reaction yields only 70 to 80% of impure end product due to a variety of side reactions. For example, in the chlorination of ethylene carbonate in substance or solution, 2-chloroacetaldehyde, polychlorinated ethylene carbonate and chlorinated ring-opening products are formed besides others. The separation of the by-products from the final product by distillation by thin-film evaporator, fractional recrystallization or zone melting is very expensive. The content of by-products can be reduced by stirring with sodium borohydride or urea at elevated temperature. However, the purification is complicated by the pronounced thermolability of vinylene carbonate, as it decomposes at temperatures above 80 °C within minutes. Highly pure vinylene carbonate can be obtained in yields of more than 70% by optimizing the chlorination conditions to suppress the formation of by-products and a combination of several gentle purification processes. The tendency of the liquid vinylene carbonate to polymerize is suppressed by addition of inhibitors such as butylhydroxytoluene (BHT).

Solid material is normally kept frozen at about -20 degrees Celsius, desiccated and protected from light. Solutions are held cold and used within a defined window because degradation products accumulate over time.
Mass spectrometry, usually electrospray ionisation coupled to liquid chromatography, gives an observed mass for comparison with the theoretical value. Peptide mapping after digestion provides complementary sequence-level confirmation.
Short exposure during weighing and handling is generally tolerated, but prolonged storage above refrigerated conditions raises the risk of deamidation, oxidation, and aggregation. The rate depends on pH, buffer composition, and concentration.
It is a synthetic peptide and a dual agonist of two incretin receptors. It is not a small molecule, and it is not structurally related to the older single-receptor peptide agonists.
