Analytical characterization of tirzepatide typically employs reversed-phase high-performance liquid chromatography (RP-HPLC) for purity assessment and peptide mapping. Mass spectrometry, often coupled with electrospray ionization, confirms molecular weight and sequence integrity. Amino acid analysis and capillary electrophoresis may also be used to detect impurities or degradation products. These methods are essential for batch release and stability studies.
Storage recommendations for tirzepatide generally specify refrigeration at 2–8 °C to maintain stability. The peptide should be protected from light and kept in its original packaging to prevent aggregation or adsorption. Freezing is not recommended because freeze-thaw cycles can cause aggregation or precipitation. Once dispensed, storage conditions and in-use periods follow product-specific labeling, which may allow room temperature storage for a limited time.
Degradation pathways for tirzepatide include deamidation, oxidation, and aggregation, which are common for therapeutic peptides. These processes can be monitored by size-exclusion chromatography (SEC) for aggregates and ion-exchange chromatography for charge variants. Forced degradation studies under acidic, basic, oxidative, and thermal stress help identify potential impurities. The exact stability profile depends on formulation, concentration, and container-closure system.
The molecule is a synthetic 39-amino-acid peptide whose backbone derives from the sequence of human glucose-dependent insulinotropic polypeptide, with several substitutions that raise metabolic stability and shift receptor preference. A C20 fatty diacid is attached through a short linker to a lysine side chain, a modification that increases binding to serum albumin. The reported monoisotopic mass is approximately 4813 Da. Near neutral pH the peptide carries a net negative charge, and the lipid tail makes the molecule markedly more hydrophobic than the unmodified parent sequence.
Dual agonism at the GIP and GLP-1 receptors underlies the observed pharmacology. Activation of GLP-1 receptors raises glucose-dependent insulin release, lowers glucagon secretion, slows gastric emptying and reduces appetite. GIP receptor activation contributes additional effects on adipose tissue and on energy balance, and the combined action on appetite appears larger than either pathway alone in animal models. Signalling bias and the relative contribution of each receptor arm to weight-related effects remain areas of active investigation.

Pharmacologically, tirzepatide activates two distinct G protein-coupled receptors: the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. Binding at each target triggers cyclic AMP accumulation and downstream signaling in pancreatic beta cells, adipose tissue and the central nervous system. Because the two pathways overlap only partially, the combined effect on insulin secretion, glucagon suppression and appetite signaling differs from that of selective single-receptor compounds. Affinity is not equal across the two targets, and the clinical meaning of that imbalance remains an area of active study.
Clinical research programs have evaluated tirzepatide in adults with type 2 diabetes and in adults with obesity or excess weight. Trials generally reported reductions in glycated hemoglobin and body weight across treatment periods of several months. Since these studies enrolled defined populations under controlled conditions, the findings describe group averages rather than individual outcomes. Open questions include the durability of effects after treatment stops, variation among subgroups, and the long-term consequences of sustained dual receptor stimulation. Published trial summaries should be consulted for exact measurements rather than secondary accounts.
An extended fatty diacid moiety promotes binding to serum albumin, which slows renal clearance and extends the circulating half-life to roughly five days. That property supports once-weekly administration and largely explains the dosing interval described in clinical reports. Published data come mainly from large randomised programmes that evaluated glycaemic control and body weight over periods of many months. Long-term outcomes beyond those trial windows, including what happens after treatment stops, remain an active area of investigation.
Tirzepatide is a synthetic peptide built from thirty-nine amino acids. Its sequence is derived from native glucose-dependent insulinotropic polypeptide, or GIP, with several non-natural residues and a fatty diacid side chain attached through a linker. The molecule behaves as a dual agonist at two incretin receptors, GIP and GLP-1, instead of targeting a single receptor. This dual engagement separates it from earlier single-receptor incretin compounds and underpins most of its reported pharmacological activity.
In a 2014 paper detailing the experimental results of the chemical characterization of flerovium, the GSI group wrote: "[flerovium] is the least reactive element in the group, but still a metal." Nevertheless, in a 2016 conference about chemistry and physics of heavy and superheavy elements, Alexander Yakushev and Robert Eichler, two scientists who had been active at GSI and FLNR in determining flerovium's chemistry, still urged caution based on the inconsistencies of the various experiments previously listed, noting that the question of whether flerovium was a metal or a noble gas was still open with the known evidence: one study suggested a weak noble-gas-like interaction between flerovium and gold, while the other suggested a stronger metallic interaction. The longer-lived isotope 289Fl has been considered of interest for future radiochemical studies. Experiments published in 2022 suggest that flerovium is a metal, exhibiting lower reactivity towards gold than mercury, but higher reactivity than radon. The experiments could not identify if the adsorption was due to elemental flerovium (considered more likely), or if it was due to a flerovium compound such as FlO that was more reactive towards gold than elemental flerovium, but both scenarios involve flerovium forming chemical bonds.
On 24 March, Israel or its allies shot down an Iranian missile crossing above Lebanon at high altitude, according to the Lebanese army, which also confirmed that the missile was not targeting Lebanon. This was the first time during the current conflict when a shootdown of an Iranian missile took place over Lebanese territory. A US strike on the Habbaniya military base—which hosts units from the Popular Mobilization Forces—in Al Anbar Governorate killed seven members of the Iraqi Armed Forces and injured 13 others. In response, Iraqi prime minister Mohammed Shia' al-Sudani summoned the US embassy's chargé d'affaires in Baghdad. Iranian state media threatened that Iran could seize Bahraini and Emirati territory if the US "makes any mistakes", while signaling its readiness to open a new front in Yemen by closing the Bab al-Mandab Strait together with the Houthis. Israel said it killed Iran's top naval commander, Alireza Tangsiri, purportedly as part of an effort to reopen the Strait of Hormuz, which Katz said Tangsiri was "directly responsible" for the closure of. On 27 March, a day after Trump and Hegseth announced the "neutralization" of Iran's military, an Iranian missile and drone strike on Prince Sultan Air Base damaged several US refueling aircraft and injured at least 15 US soldiers. An E-3 Sentry was also damaged by the strike. Military analyst Cedric Leighton described the attack as a "a serious blow to [US] surveillance capabilities". The same day, Iran blocked two Chinese ships from entering the strait and sought to formalize fees for ships passing through it.
===== Cetacean research ===== Peacock & Thewissen (2026) compare volume measurements of the bony labyrinth in extant mammals and Eocene cetaceans, and hypothesize that fossil cetaceans had membranous ducts of relatively larger size compared to terrestrial even-toed ungulates, and that vestibular organs of early cetaceans were necessary for terrestrial locomotion. Buono et al. (2026) reconstruct changes of cetacean morphological diversity and ecospace occupancy throughout the evolutionary history of the group, identifying Eocene–Oligocene and Plio–Pleistocene transitions as periods of major evolutionary changes, and linking changes of ecospace occupancy to the main radiation events of the group. Tyborowski et al. (2026) report the discovery of a portion of a dentary of an indeterminate member of Pelagiceti from the Siemień Formation, representing the first record of an Eocene whale from Poland and one of the smallest fully aquatic Eocene whales reported to date. A deciduous tooth of a basilosaurid with evidence of malformation that might have been caused by localized stress is described from the Eocene Submeseta Formation (Seymour Island, Antarctica) by Bajor et al. (2026). Hakkens, Reumer & Schulp (2026) study the affinities of the basilosaurid specimen TM 8501 from the Eocene (Priabonian) Yazoo Clay (Alabama, United States; the holotype of Zeuglodon hydrarchus), identify the studied individual as a mid-sized cetacean distinct from Dorudon serratus and Chrysocetus healyorum, an tentatively refer the specimen to Zygorhiza kochii. The white shark is an obligate ram ventilator; to breathe, it must swim constantly so water flows through its gills. Over 95% of the shark's musculature is white, fast-twitch muscles, which allows it to move in quick sprints, particularly when ambushing prey. The remaining dark, slow-twitch muscles carry oxygen to power the shark while at cruising speed. The white shark has a large, double-lobed liver that can be almost 30% of its body weight and stores lipids, fatty acids, and oils. The liver helps keep the shark from sinking, as the oil is six times more buoyant than the surrounding water. The lipids and fatty acids provide the shark with energy for travel and are important for reproduction and growth. One study concluded that a white shark liver is more energy-rich than whale blubber. White sharks appear to have strong immune systems and can tolerate high amounts of toxic heavy metals like mercury and arsenic in their blood, more so than many other vertebrates. They are also documented to heal relatively quickly from even severe wounds, and the species's genome shows "positive selection in key genes involved in the wound-healing process". White sharks use a system of blood vessels to warm up portions of their body (regional endothermy), which allows them to be active and hunt in cool waters. In this complex blood vessel system, known as a rete mirabile, warm blood generated from the dark muscles is constantly supplied to other parts of the body.
The secondary stage occurs months to two years later (but usually 1–2 months later), and may thus begin when the mother wound has not yet healed. It happens when the bacterium spreads in the blood and lymph. It begins as multiple, pinhead-like papules; these initial lesions grow and change in appearance and may last weeks before healing, with or without scarring. Secondary yaws typically shows widespread skin lesions that vary in appearance, including "crab yaws" (areas of skin of abnormal color) on the palms of the hands and soles of the feet (named for the crab-like gait they cause people with painful soles to assume). These may show desquamation. These secondary lesions frequently ulcerate and are then highly infectious, but heal after 6 months or more. Secondary yaws affects the skin and bones. The most common bone-related problem is periostitis, an inflammation around the bone, which often occurs in the bones of the fingers and the long bones of the lower arms and legs, causing swollen fingers and limbs. This causes pain at night and thickening of the affected bones (periostitis). About 75% of infected children surveyed in Papua New Guinea reported joint pain. Swollen lymph nodes, fever, and malaise are also common. A latent infection develops after primary and secondary yaws (and possibly, in some cases, without these phases). Within five years (rarely, within ten years), it can relapse and become active again, causing further secondary lesions, which may infect others. These relapse lesions are most commonly found around the armpits, mouth, and anus.
His contemporary Alfred Russel Wallace also noted these variations and the geographical separations between different forms leading to the study of biogeography. Wallace was influenced by the work of Philip Lutley Sclater on the distribution patterns of birds.
=== Whipple's triad === The symptoms of low blood sugar alone are not specific enough to characterize a hypoglycemic episode. A single blood sugar reading below 70 mg/dL is also not specific enough to characterize a hypoglycemic episode. Whipple's triad is a set of three conditions that need to be met to accurately characterize a hypoglycemic episode. The three conditions are the following:
Anabolic effects include growth of muscle mass and strength, increased bone density and strength, and stimulation of linear growth and bone maturation. Androgenic effects include maturation of the sex organs, particularly the penis, and the formation of the scrotum in the fetus, and after birth (usually at puberty) a deepening of the voice, growth of facial hair (such as the beard) and axillary (underarm) hair. Many of these fall into the category of male secondary sex characteristics. Testosterone effects can also be classified by the age of usual occurrence. For postnatal effects in both males and females, these are mostly dependent on the levels and duration of circulating free testosterone.
